Abstract:

The aim of this research was to develop a simple, selective and sensitive aptasensor for detection of Pb2+ based on hairpin structure of complementary strand (CS) of aptamer (Apt). Screen printed carbon electrode (SPCE) was electrodeposited by gold nanoparticles to be used as the substrate for the immobilization of Apt-CS. Moreover, gold nanoparticles (AuNPs) and thionine were added to increase the sensitivity of the aptasensor. The morphology of SPCE and AuNPs modified SPCE was investigated using scanning electron microscopy (SEM). The SEM results showed that the AuNPs were homogeneously distributed on the surface of SPCE. The electrochemical performance of aptasensor was evaluated by means of cyclic voltammetry (CV) and differential pulse voltammetry (DPV) measurements. The electrochemical results indicated that peak current due to the hairpin structure formation of CS decreased in the presence of Pb2+. However, in the absence of Pb2+, peak current was enhanced because of the combination of thionine/AuNPs-CS. The designed aptasensor showed a high selectivity toward Pb2+, wide linear range (1-40 nM), low detection limit (374 pM), acceptable reproducibility and good long-term stability.

An Electrochemical Sensor Based on Novel Ion Imprinted Polymeric Nanoparticles for Selective Detection of Lead Ions

In this study, the novel surface ion-imprinted polymer (IIP) particles were prepared and applied as a electrode modifier in stripping voltammetric detection of lead(II) ion. A carbon paste electrode (CPE) modified with IIP nanoparticles and multi-walled carbon nanotubes (MWCNTs) was used for accumulation of toxic lead ions. Various factors that govern on electrochemical signals including carbon...

Electrochemical aptasensor for tetracycline detection.

An electrochemical aptasensor was developed for the detection of tetracycline using ssDNA aptamer that selectively binds to tetracycline as recognition element. The aptamer was highly selective for tetracycline which distinguishes minor structural changes on other tetracycline derivatives. The biotinylated ssDNA aptamer was immobilized on a streptavidin-modified screen-printed gold electrode, a...

Development of an aptasensor for electrochemical detection of tetracycline

Developing the rapid, simple and sensitive biosensor system for tetracycline detection is very important in food safety. In this paper, we developed a label-free aptasensor for electrochemical detection of tetracycline. The reorganization of tetracycline binding aptamer was confirmed by Isothermal Titration Calorimetry, Kd 1⁄4 5.18 10 5 mol L 1. According to the electrochemical impendence spect...

Development of an aptasensor for electrochemical detection of exosomes.

Exosomes are small (50-100 nm in diameter) vesicles secreted from various mammalian cells. Exosomes have been correlated with tumor antigens and anti-tumor immune responses and may represent cancer biomarkers. Herein, we report on the development of an aptamer-based electrochemical biosensor for quantitative detection of exosomes. Aptamers specific to exosome transmembrane protein CD63 were imm...

Label-Free Aptasensor for Lysozyme Detection Using Electrochemical Impedance Spectroscopy

This research develops a label-free aptamer biosensor (aptasensor) based on graphite-epoxy composite electrodes (GECs) for the detection of lysozyme protein using Electrochemical Impedance Spectroscopy (EIS) technique. The chosen immobilization technique was based on covalent bonding using carbodiimide chemistry; for this purpose, carboxylic moieties were first generated on the graphite by elec...
